# Polygenic score for osteoporosis shows modest discrimination gain in Japanese adults

Researchers developed a polygenic score (PGS) for osteoporosis using heel ultrasound T-scores in up to 10,794 Japanese participants and validated it in 8,711 independent individuals. Adding the PGS to age and sex produced only modest gains in discrimination, but quintile-based stratification showed a gradient: individuals in the lowest genetic risk quintile had 42% higher incidence of osteoporosis over 3.5 years, while those in the highest quintile had 30% lower incidence. The effect was similar at baseline and during follow-up, and did not vary by age.

*Why it is interesting: Tests whether polygenic prediction of a common complex disease can offer clinically useful stratification in a non-European ancestry population and whether genetic risk is detectable before peak bone mass.*
