Individualised mRNA melanoma vaccine with pembrolizumab cuts recurrence or death by 42%

Adding intismeran autogene to pembrolizumab after melanoma was removed reduced recurrence or death risk by 42% in phase III INTerpath-0001.

This news item reports results of the phase III INTerpath-0001 trial in patients whose melanoma had been surgically removed. The trial added intismeran autogene, an individualised mRNA neoantigen vaccine, to the PD-1 inhibitor pembrolizumab. The comparison was against adjuvant pembrolizumab alone. The combination reduced the risk of disease recurrence or death by 42%. The results were presented at the ESMO Congress 2026 in Madrid. The abstract gives no sample size, follow-up time, confidence interval or overall survival data.

Why it is interesting: An individualised mRNA vaccine shows a 42% reduction in recurrence or death in a phase III adjuvant trial. The vaccine is built from each patient's own tumour neoantigens.
